Study of Morphometry on Langerhans' Islet Endocrine Cells in Obese Rats Induced by Means of High Fat Diet



Abstract
This study aimed to evaluate the changes of Langerhans' islet and its endocrine cells in pancreas of obese rat induced by intake of high fat diet. The glucagon, insulin, and somatostain granules in A, B and D cells of pancreas were stained by
immunohistochemical method at 4, 8 and 12 weeks intervales after high fat diet And the area of Langerhans' islet and granules were measured by computer image analyzer, the area percentage of each granules of obese rats was calculated and with
those
of
lean rats.
@ES The final outcomes were obtained as follows ;
@EN 1. In changes of body weight, weight, increased percentage of body weight was 3.4% at 4 weeks, 13.9% at I weeks, 21.5% at 12 weeks in high fat diet group as compared to control lean group.
2. In distributions of number of Langerhans' islet classified by size, the number of small sized islets were decreased & the number of large sized islets were increased in high fat diet group at 8 and 12 weeks as compared to control lean group,
but
total numbers of observed islets in each group were not different statistically.
3. In area of Langerhans' islet, significant increase was observed at 8 and 12 weeks in high fat diet group, and its increased percentage was 12.4% at 8 weeks and 23.4% at 12 weeks as compared to control lean group.
4. In area of glucagon granules, significant decrease was observed at 12 weeks in high fat diet group and its decreased percentage was 9.5% as compared to control lean group.
5. In area percentage of glucagon granules, significant decreased was observed at 8 and 12 weeks in high fat diet group, and its decreased percentages was 13.3 0.6% at 8 weeks and 12.4 0.6% at 12 weeks compared with 16.0 0.7% and 16.8 0.8% of
control
lean group.
6. In area of insulin granules, significant increase was observed a 4 weeks in high fat diet group, and its increased percentage was 12.6% as compared to control lean group.
7. In area percentage of insulin granules, significant decrease was observed at 12 weeks in high fat diet group, and its decreased percentage was 57.7¡¾2.4% compared with 70.2¡¾2.1% of control lean group.
8. In area and area percentage of somatostatin granules, no significant change was observed in high fat diet group as compared to control lean group.
